Add that price in your listing for list price and have your regular price be same and keep it for few days so you can have sales at the higher price. Lower your selling price and now it should show strike through.
I have a price of 48.99 and 20% off. Does this off percentage count?

Have you sold at that price? Also the 20% as a coupon? That is something else. The strike through price is a lower price than normal not a coupon

I sold at 48.99-20%=39.19. So should I do the price lower than that?

The strike through is something else. So if u want that you have to first update list price and make your price same $48.99 removing discount until u make few sales and then change your price to $39.19

You must need to put a higher amount in the list price and then sell a few units at that price and later on then put the sale price, this will help you to get that strikethrough price.
